Juve won their first UEFA Champions League match in three years with a 4-0 drubbing of FC Nordsjaelland on Wednesday night.
The win left them on six points in Group E - one point behind joint-leaders Chelsea and Shakhtar Donetsk.
The Bianconeri next face the Blues and will be looking to leapfrog the team they drew 2-2 with last time out courtesy of a last-gasp Arturo Vidal equaliser.
Buffon knows the task ahead is not an easy one against the defending European champions and realises the time to shine is now.
Speaking after the midweek victory, the 34-year-old shot-stopper said: "I got the gloves dirty, but it was a standard save. If I didn't make it I should think about retiring.
"Little has changed in reality. We Italians are inclined to look at the results of others and consider them to be more important than our own.
"Instead, the good thing is that we're two games away from the end of the group stage and our destiny remains in our hands.
"If we do well we'll go through, otherwise we'll play in the Europa League.
"Chelsea have almost a decade's worth of experience in playing these types of games.
"Shakhtar, with their individual brilliance combined with their team play, are more unpredictable and a tough prospect to face."
