Mourinho's decision to once again pick the Madrid-born shot-stopper ahead of club captain Iker Casillas was greeted with whistles and boos from the home supporters.
The Portuguese tactician feels the fans' reaction shows complete disrespect towards Adan.
"If the fans whistle me because they do not agree with my decision and the team play badly, then I accept it," Mourinho said at a post-match press conference.
"I spoke to Adan prior to the match and told him the whistles were for me and not for him. But in my opinion, that (the whistles) showed a real lack of respect for him.
"This is a player who has been at Real since he was 16, who was born in Madrid and who does not deserve that."
Casillas' arrival on the pitch after Adan was sent-off in the sixth minute was applauded by the Madridistas.
This latest chapter has emphasized the ever-increasing rift that has developed between Mourinho and Real fans, and further fueled speculation that his departure at the end of the season is inevitable.
