The Red Devils went ahead with only 36 seconds on the clock as Van Persie turned his defender and hit a shot which was on target but took a lucky deflection off James Collins to loop over Jussi Jaaskelainen into the net.
Kevin Nolan flicked a free kick into the side netting as West Ham looked to come back into it, while Anderson and Javier Hernandez brought good saves out of Jaaskelainen before half-time.
In the second half, Tom Cleverley flashed a shot just over the bar on 50 minutes while Van Persie hit a free-kick straight at the keeper and Rooney saw a shot saved brilliantly as United dominated for long periods.
But Carlton Cole almost grabbed a point for the Hammers on 81 minutes as a deep free kick landed at his feet and United needed Anders Lindegaard to get down well to stop his shot and ensure United held onto all three points.
