The England international fired home all three goals as Spurs earned a 3-1 victory over Maribor to take the north London club second in Group J with two games remaining.
The treble took Defoe to 126 career goals the club, two more than White Hart Lane legend Sheringham, and he admits he is delighted to have achieved such a milestone.
"It's special to go past Teddy," Defoe, who is now eighth in Tottenham's all-time goal scoring charts, told ITV.
"He was a great player for club and country. I watched him when I was growing up so it's great for me personally."
On what the result meant for his club, Defoe added: "We needed to win to bounce back from the weekend [defeat to Wigan].
"We pressed high from the minute go. The tempo was good, the movement was good, we kept the ball better and we're all delighted."
