Harambee Stars midfielder David Ochieng scored twice and Michael Olunga and debutant Andrew Segkayombya netted a goal each to give the visitors a comfortable footing ahead of the second-leg.
The return leg will be played in Nairobi in two weeks time and Tusker are likely to face Egyptian giants Al-Ahly in the next round.
