Issam Jemma scored twice to give the Carthage Eagles a comfortable lead heading into the break, but it was an all Black Stars affair in the second period as Albert Adomah, John Boye, Mubarak Wakaso and captain Asamoah Gyan found the back of the net.
The game was played behind closed doors as both teams preferred not to give away too much about their respective game plans ahead of the tournament, which kicks off on Saturday, and for Ghana, who last won the title in 1982, the result came as a timely boost ahead of their trip to South Africa.
Ghana are in Group B with the Democratic Republic of Congo, Mali and Niger and will play all their games in Port Elizabeth at the Nelson Mandela Bay Stadium, with their first game coming on Sunday against DR Congo.
Tunisia, meanwhile, are in Group D with Ivory Coast, Togo and Algeria and they face the latter next Tuesday at the Royal Bafokeng Stadium in Rustenburg.
