Super Eagles coach Stephen Keshi had named both players in his squad for the midweek trip to the United States as he continues his preparations for the 2013 Africa Cup of Nations.
However, Nigerian FA officials confirmed on Monday that midfielder Mikel and forward Moses will not be required for duty after "requesting time off to either deal with a personal issue and recover from a minor knock".
Mikel was included in Roberto Di Matteo's starting line-up against Liverpool on Sunday and played the entire 90 minutes, while Moses was introduced as a second-half substitute during the 1-1 draw at Stamford Bridge.
Meanwhile, Newcastle striker Shola Ameobi has been given the chance to impress for the African giants after receiving his maiden call-up for the country of his birth.
Keshi admitted Ameobi could stake his claim for a place in the coach's AFCON squad if he delivers against the South Americans.
"He brings something special, a different dimension," Keshi told the BBC. "This friendly is an opportunity for some players who have not featured in our recent games to show what they can do ."
