This, after the defender was a notable absentee in Chiefs' back-line for Saturday's 2-0 defeat to Dikwena at the Royal Bafokeng Stadium.
It was Amakhosi's first loss of the Premiership season, although Stuart Baxter's men remain top of the league standings.
"Mathoho has got a strained muscle in his buttock," Milner told Chiefs' official website.
"We were concerned he may aggravate it against Platinum Stars so had to keep him out. He has now started jogging."
The Soweto outfit are still without the services of Morgan Gould, Jimmy Tau and Jimmy Jambo due to long-term injuries, with Milner keen to update the Glamour Boys' fans on their stars' recoveries.
He explained: "He (Gould) is recovering nicely and is still on track to return in February. He is working hard at his rehab.
"(Tau) will be seeing a doctor this week and will be assessed. He is still expected back in July of 2013.
"(Jambo) is still in braces and will be out for three months. He will start his rehab programme this week and is expected back in a year's time."
